Pharmacological Properties:

Pharmacodynamics: Cefpodoxime proxetil is a broad-spectrum third-generation cephalosporin antibiotic for oral use. After absorption through the gastrointestinal tract, it is hydrolyzed by enzymes into cefpodoxime to exert its antibacterial effect. It is effective against both Gram-positive and Gram-negative bacteria. The mechanism of action of this product is to achieve a bactericidal effect by inhibiting the synthesis of the bacterial cell wall. Susceptible bacteria mainly include Staphylococcus, Streptococcus, and Gram-negative bacteria (such as Pasteurella, Proteus, and Escherichia). This product has no antibacterial activity against most obligate anaerobes (such as Pseudomonas or Enterococcus).

Pharmacokinetics: After oral administration, cefpodoxime proxetil is absorbed through the gastrointestinal tract and de-esterified in the body to form active cefpodoxime, which enters the systemic circulation. In Beagle dogs administered orally at a dose of 10 mg/kg body weight, the time to peak plasma concentration of cefpodoxime is 2.52 ± 1.21 hours, the peak plasma concentration is 19.03 ± 13 μg/mL, the elimination half-life is 10.46 ± 4.69 hours, the clearance rate is 69.76 ± 22.75 mL/h/kg, and the relative bioavailability is 97.49%.

Drug Interactions: Concomitant use with antacids or H2 receptor antagonists may decrease the absorption and plasma concentration of cefpodoxime.

Indications: 

Used for the treatment of skin infections (wounds and abscesses) in dogs caused by Staphylococcus aureus, Staphylococcus intermedius, Streptococcus canis, Escherichia coli, Pasteurella multocida, and Proteus mirabilis.

Dosage and Administration: 

Calculated by cefpodoxime. Oral administration: 5–10 mg per 1 kg of body weight for dogs. Once daily, for 5 to 7 consecutive days, or until 2 to 3 days after clinical symptoms have resolved, up to a maximum of 28 days.

Adverse Reactions: Occasional vomiting, diarrhea, polydipsia, and decreased appetite.

Precautions:

Contraindicated in dogs with known hypersensitivity to cefpodoxime or β-lactam (penicillin and cephalosporin) antibiotics.

Antibacterial drugs, including penicillins and cephalosporins, can cause allergic reactions in sensitive individuals. To minimize the potential for allergic reactions, avoid direct contact of skin and mucous membranes with this product when handling.

The safety of cefpodoxime proxetil in breeding, pregnant, or lactating dogs has not been established.

Like other cephalosporins, cefpodoxime proxetil has been rarely associated with a positive direct Coombs' test.

Keep out of reach of children.

When treating systemic infections, if no therapeutic effect is observed after 3 to 4 days of consecutive use, this product should not be continued.

The dosage should be determined based on factors such as the severity and nature of the skin infection in the affected dog, the susceptibility of the pathogenic microorganisms, and the integrity of the host defense mechanisms of the affected dog.

Fasting is not required before or after administration.

Withdrawal Period: No withdrawal period needs to be established.
